Verbena officinalis, water and water-alcohol extractsAbstract
Introduction. An important task is to search pharmacy and new herbal medicines. Phytomedication characterized by a wide spectrum of pharmacological action and low toxicity, they can be used for a long time to prevent and treat diseases. Widespread use of herbs and drugs based on them, due to the presence of different chemical composition and action of biologically active substances that have the ability to put it included in the metabolism and cause minimal side effects. Flora of Ukraine is characterized by diverse species composition of plants. Particularly noteworthy plants from centuries of experience in the use of traditional medicine, which include species of Verbena [1, 2].
Verbena officinalis widespread in Ukraine, exhibits anti-inflammatory, antimicrobial and hepatoprotective effect [3, 4].
The purpose of these studies was to obtain safe water and definitions (VLT-0) and the water-alcohol (VLT-7) extracts from medicinal herb verbena, establish the pharmacological activity of investigated extracts.
Materials and methods
The objects of study chosen extracts verbena (v.). The Department of Pharmacy Ivano-Frankivsk National Medical University obtained extracts from the studied material.
The main task of grinding raw materials can be considered destruction of cell structure and increased surface extraction [5, 6]. Type extractant affects not only the extraction of specific substances, but the total amount of extractives depend on the hydrophilic extractant. Given that many substances in plants relates to hydrophilic, polar extractants will extract more extractives [6].
Alcohol extracts of small ethanol (40%) contain a lot of high-molecular compounds (water-soluble proteins, sugars, enzymes, pectins, mucus, starch) that should be removed [6].
Due to the chemical composition of medicinal herb verbena, us as extractant used 70% ethanol and purified water.
The starting material for obtaining extracts were dried herb verbena (humidity 6 - 7%), ground to a particle size of 1 - 3 mm, which is harvested in 2014.
For aqueous extract 200g chopped plant material extracted with 2.0 liters of purified water in a flask with reflux in a boiling water heater for 30 minutes. The extract was filtered, and the residue of raw materials extracted in similar conditions 2 more times. Extracts all filtered and filtrates combined.
For aqueous alcoholic extract of herbs 200g chopped plant material extracted with 200 ml of 70% ethanol in a flask with reflux at the boiling point ektrahentu. The obtained extracts were combined, ethanol distilled under vacuum. After stripping the ethanol extract total volume of purified water adjusted to the original volume.
Results and discussion
As a result, the study found that aqueous and aqueous-alcoholic extracts of herbs Verbena officinalis in a single intra-gastric administration at a dose of 5000 mg / kg for the entire observation period the animals were kept normal reflexes, abnormalities in behavior, clinical symptoms of intoxication and death of animals was not found which indicates no significant toxic effects of extracts of Verbena officinalis.
Past studies indicate anti-inflammatory activity, among all investigated extracts the highest anti-inflammatory activity manifests aqueous alcoholic extract of herbs Verbena officinalis.
The study of hepatoprotective activity of extracts of medicinal herb verbena conducted at screening model of liver damage that occurs when injected carbon tetrachloride. Established that the death rate in the control group was 16.7% in the other groups all animals remained alive until the end of the experiment.
Results antimicrobial studies suggest that extracts retard growth , which are often agents of hospital infections by pathogens and secondary infection. Most sensitive to the action of the extracts are strains of Staphylococcus, and the most resistant - strains of Pseudomonas aeruginosa.
Conclusions
1. Оbtain water and aqueous-alcoholic extracts of herbs Verbena officinalis.
2. Study of acute toxicity showed that water and water-alcohol extract of medicinal herb verbena is non-toxic substances in the intra-gastric administration.
3. Еstablished that extracts exhibit antimicrobial, anti-inflammatory and hepatoprotective activity. Water-alcohol extract of Verbena officinalis is a promising substance for the development of drugs with hepatoprotective effect.
