CHROMATOGRAPHIC STUDY OF THE COMPONENT COMPOSITION OF THE PHENOLIC COMPOUNDS OF THE WOOD COW-WHEAT (MELAMPYRUM NEMOROSUM L.)

Abstract

The aim of the work - to determine the component composition of flavonoids, phenolic acids and catechins in the Wood Cow-wheat (Melampyrum nemorosum L.) herb collected in Ukraine.

Materials and Methods. The herb of M. nemorosum was harvested in 2022 in the flowering phase near the village. Mykytsy residents of Kosiv district, Ivano-Frankivsk region. The component composition of phenolic acids, flavonoids, and catechins was determined by the method of high-performance liquid chromatography (HPLC) on an "Agilent Technologies 1200" liquid chromatograph equipped with a flow-through vacuum degasser, a four-channel low-pressure gradient pump, an autosampler, a column thermostat, and a diode array detector, using Zorbax SB-C18 columns and Zorbax SB-Aq.

Results and Discussion. Using HPLC, it was established that the herb of M. nemorosum contains 9 flavonoids (rutin, quercetin-3-b-glucoside, naringin, neohesperidin, quercetin, naringenin, apigenin, rhamnetin, kaempferol); among them 5 flavonols, 3 flavonones and 1 flavone were identified. There were also revealed 3 catechins (catechin, epicatechin, and gallocatechin), and 9 phenolic acids (hydroxyphenylacetic, chlorogenic, caffeic, syringic, benzoic, p-coumaric, trans-ferulic, sinapic, and trans-cinnamic).

Conclusions. 1. The component composition of flavonoids, phenolic acids and catechins in the herb M. nemorosum harvested in Ukraine was determined using HPLC. 2. Among the flavonoid compounds, rutin (3388.58 μg/g), quercetin (716.44 μg/g), naringin (679.21 μg/g), rhamnetin (662.69 μg/g), and neohesperidin (588 .90 μg/g). Among the phenolic acids, benzoic acid dominates (948.54 μg/g), and among catechins, epicatechin (415.71 μg/g). 3. A promising direction of further research is the study of the component composition of phenolic compounds of the M. nemorosum herb collected in different regions of Ukraine.
