CYTOLYSIS MARKER INDICATORS IN RATS AFFECTED BY HIGH DOSES OF ADRENALINE IN THE DYNAMICS OF POISONING WITH THE FOOD DYE AZORUBINE

Abstract

SUMMARY. Today, synthetic food dyes are widely used in the food industry – water-soluble organic compounds that do not occur in nature. They do not have their own nutritional, energy or other value, but are used in food products exclusively to give them an attractive appearance and various colors. At the same time, the results of modern studies increasingly indicate their possible negative impact on the body. Among people who consume such products are those who suffer from cardiovascular diseases. One of the key pathogenetic factors in the development of such pathology is the excessive intake of catecholamines, which is observed with intense or frequent stress. The combination of the use of products with an excess of dyes, as well as stressful situations that are common today, pose a threat to human health.

The aim – to determine the activity of cytolytic processes in rats affected by elevated doses of adrenaline, against the background of poisoning with the food dye azorubine.

Material and Methods. The studies were conducted on 78 white male rats, which were kept on a standard diet of the vivarium of the Ternopil National Medical University. The animals were divided into 13 groups of 6 individuals each, with one group serving as an intact control. Dye E122 was administered intragastrically daily for 21 days at a dose of 100 mg/kg body weight. Acute adrenaline myocardial damage was induced by a single intramuscular injection of 0.18 % adrenaline hydrotartrate solution at a dose of 0.5 mg/kg. Euthanasia was performed using sodium thiopental at 3, 24 and 48 hours after adrenaline administration against the background of azorubine poisoning after 7, 14 and 21 days. In the blood serum, heart and liver of experimental rats, the activity of alanine aminotransferase and aspartate aminotransferase, gammaglutamyl transpeptidase, lactate dehydrogenase was determined, and in whole blood, the erythrocyte intoxication index was determined.

The results were subjected to statistical processing using the STATISTICA 13 program.

Results. In the blood serum of rats affected by adrenaline on the background of azorubine poisoning, a progressive increase in aminotransferase activities was observed, which at the end of the experiment was the highest (ALT increased 3.4 times compared to normal and 1.2 times compared to rats poisoned only with azorubine). A similar increase was observed in the activity of AST, GGTP and LDH, a decrease in the activity of these enzymes was observed in the liver and myocardium, and with the extension of the study period it was more pronounced. When studying the percentage of erythrocyte membrane permeability, its increase was established at all times of the experiment. Within 21 days of azorubine poisoning and 48 hours of adrenaline exposure, the permeability of the erythrocyte membrane increased by 69 % compared to this indicator in intact control rats, and compared to rats poisoned with azorubine during the same period, the indicator increased by 8 %.

Conclusions. An additive effect of the action of the food dye azorubine and adrenaline on the rat body was established, which is confirmed by the results obtained by us from the study of cytolysis marker indicators.
