REGULARITIES OF ERYTHROCYTE, PLATELET AND LEUKOCYTE INDICES AND PLASMA ENZYMES IN DIFFERENT AGE GROUPS OF INTACT RATS

Abstract

The aim of the work. To identify the peculiarities of changes in hematological parameters of blood in the age aspect.

Materials and Methods. The study was conducted on 15 outbred Wistar male rats that were divided into three groups of 5 animals by age, group I – immature, 1.5–2 months of age, weighing 80–120 g, group II of sexually mature rats, 6 months, weighing 180–200g and old rats in group III with a weight of 320–360g, aged 20 months. For the study, whole blood and serum were selected, blood was collected from the heart cavity. The study of the parameters of general blood analysis was performed on HoribaYumizenH500 hematological analyzer, and the evaluation of the activity of blood plasma enzymes on the biochemical analyzer Master T.

Results and Discussion. On the basis of the analysis of the biochemical profile of blood plasma in the group II, compared to the III, a significant increase of ASAT and AlAT was found by 36.8 and 3.27 %, respectively, and in the group I compared to the group III – by 94.5 % and 9.5 %. A similar trend is observed in the analysis of alkaline phosphatase (AP) so that in the group I revealed a significant increase in its activity by 202.04 % compared with the indicator in the group III, and in the group II compared to the III – by 98.5 %. Analysis of the width of red blood cell distribution showed that in group I compared with the RDW-CV and RDW-SD, significantly increased by 8.83 % and 9.77%, respectively, and compared with the group III by 13.1% and 15.73%. A relatively small increase in MCV in group I was found compared to II MCV by 0.76 % and 4.12%, respectively. Unlike erythrocyte indices, platelet enlargement is directly proportional to the age of the animals, by groups in the sequence I (PDW, MPV, P-LCC)> II (PDW, MPV, P-LCC)> III (PDW, MPV, P-LCC) III. Analysis of two major leukocyte fractions of NEU, LYM indicates the active inclusion of the first level of immune protection in the group of mature animals. Thus, the absolute content of NEU, LYM in the group II of animals compared to the group I significantly increased by 141 % and 128.7 % respectively.

Conclusions. The increase of activity of transaminases (ALAT, ASAT), alkaline phosphatase (AP) and the increase of heterogeneity of erythrocyte fractions in the group of immature animals was revealed. Rheological analysis of platelet indices indicates an increase in the number of immature platelets and a high probability of their aggregation in the group of old rats. In general, the total value of leukocytes progressively increases as the age range increases.
