PROSPECTS OF THIAZOLE HETEROCYCLES AS INNOVATIVE ANTI-INFLAMMATORY AGENTS: A LITERATURE REVIEW

The aim of the work. To systematize current literature data on the anti-inflammatory effects of thiazole-based heterocyclic compounds, with an emphasis on their classification, mechanisms of action, structure-activity relationships, including multifunctional approaches and "prodrug" concepts.
Materials and Methods. Information search, system and content analysis, generalization of results.
Results and Discussion. Thiazole heterocycles occupy a leading position in modern medicinal chemical design due to their structural flexibility, high biological activity and the possibility of multifunctional modification. Considering the global increase in the prevalence of inflammatory diseases, as well as the limitations of traditional NSAIDs due to side effects and resistance, there is an urgent need for new anti-inflammatory agents with an increased therapeutic index. This review systematizes the current data on the classification of thiazole compounds with anti-inflammatory activity, their mechanisms of action, structure-activity relationships (SARs), as well as the results of preclinical studies in vitro and in vivo. Special attention is paid to the prospects for optimizing molecules through multifunctional approaches, "prodrug" strategies and pharmacophore hybridization.
Conclusions. Key challenges and promising directions for the development of potential anti-inflammatory agents based on the thiazole core are formulated, including the need for standardization of evaluation methods, implementation of computer modeling and artificial intelligence design. This review creates a scientific basis for the further development of thiazole anti-inflammatory agents and their integration into modern pharmacotherapy.
