ВИВЧЕННЯ ЗМІН ВУГЛЕВОДНОГО ОБМІНУ ТА МОРФОЛОГІЧНОГО СТАНУ СІТКІВКИ ОКА ЩУРІВ ЗА УМОВ ЕКСПЕРИМЕНТАЛЬНОЇ ДІАБЕТИЧНОЇ РЕТИНОПАТІЇ

Актуальність. Діабетична ретинопатія є одним із найпоширеніших мікросудинних ускладнень цукрового діабету, що призводить до прогресуючого ураження сітківки та втрати зору. Провідну роль у її патогенезі відіграють хронічна гіперглікемія та пов’язані з нею порушення вуглеводного обміну, які ініціюють структурні та функціональні зміни в органі зору. Мета – вивчити зміни показників вуглеводного обміну та морфологічного стану сітківки ока щурів за умов стрептозотоцин-індукованої діабетичної ретинопатії. Матеріали та методи. Дослідження проведено на 30 білих нелінійних щурах масою 180–200 г. Діабетичну ретинопатію моделювали шляхом одноразового внутрішньовенного введення стрептозотоцину (65 мг/кг) на тлі нікотинаміду (230 мг/кг). Оцінювали динаміку маси тіла, рівень глюкози крові та глікозильованого гемоглобіну на 60-у та 120-у добу експерименту. Морфологічні зміни сітківки вивчали гістологічними методами (забарвлення гематоксиліном і еозином). Результати. У щурів зі змодельованою діабетичною ретинопатією встановлено достовірне підвищення рівня глюкози крові та глікозильованого гемоглобіну на 60-у та 120-у добу порівняно з інтактною групою. Концентрація HbA1c на 60-у добу становила 7,6 ± 0,5 %, на 120-у – 10,4 ± 0,8 % (проти 5,8 ± 0,6 % в інтактних тварин, p < 0,05). Гістологічно на 60-у добу виявляли зміни у внутрішньому та зовнішньому ядерних шарах сітківки без ознак неоваскуляризації; на 120-у добу спостерігалося зменшення кількості біполярних клітин та поява вогнищ неоваскуляризації, характерних для проліферативної стадії ретинопатії. Висновки. Експериментальний цукровий діабет супроводжується прогресуючими порушеннями вуглеводного обміну та стадійними морфологічними змінами сітківки, що відповідають розвитку діабетичної ретинопатії від непроліферативної до проліферативної стадії.
